Investigating the Effects of Ketamine on E/I Balance using Aperiodic Activity and Computational Modeling of Human MEG-Data
Abstract
Ketamine, a NMDA receptor antagonist, produces a range of psychotomimetic effects when administered in sub-anesthetic doses. Previous research suggests that ketamine alters the excitation/inhibition (E/I) balance in cortical microcircuits by acting on inhibitory, GABAergic interneurons and thereby disinhibiting pyramidal neurons (Homayoun and Moghaddam, 2007; Seamans, 2008). However, the precise neural mechanisms, including the subtypes of GABAergic interneurons involved, are not well understood. Non-invasive measurements of gamma-band oscillations and aperiodic activity in combination with computional modelling may provide insights into the underlying mechanisms of E/I disturbances. To address these questions, we will investigate the neural effects of ketamine in humans, using MEG-data and computational modelling.
