Dimensions of Psychedelic-Induced Altered States and Their Relationship to Antidepressant Outcomes in Treatment-Resistant Depression: A Secondary Analysis of the PsiDeR Trial
Abstract
This project constitutes a secondary analysis of the NIHR-funded PsiDeR trial, a phase 2 double-blind randomised controlled trial evaluating the antidepressant efficacy of a single 25 mg psilocybin session combined with psychological support for patients with treatment-resistant depression (TRD). The current analysis aims to examine how individual dimensions (subscales) of the psychedelic experience, measured by the 5-Dimension Altered States of Consciousness Questionnaire (5D-ASC), relate to short-term antidepressant outcomes as assessed by the Montgomery–Åsberg Depression Rating Scale (MADRS) and Quick Inventory of Depressive Symptomatology–Self-Report (QIDS-SR). Hypotheses, statistical models, and covariate adjustments were formulated prior to outcome data access to ensure analytic transparency.
