Effects of NMDA-receptor blockade by ketamine on mentalizing

OSFSep 14, 2022

Abstract

Psychosis is known to be associated with deficits in meta- and social cognition (Green, Horan, and Lee, 2015). The uncompetitive NMDA-receptor antagonist Ketamine has been identified to trigger psychotomimetic symptoms in healthy adults that are similar to symptoms observed in patients suffering from schizophrenia (Curic et al., 2018). We used ketamine as a model for psychosis to investigate whether Theory of Mind is impaired in participants under the influence of Ketamine. In a randomized, double-blind, placebo-controlled between-subjects experiment, we intravenously administered ketamine or placebo to healthy participants performing a video-based social cognition task during functional magnetic resonance imaging. Psychotomimetic effects of ketamine were assessed using the Positive and Negative Syndrome Scale.
