Exploring mediators of psilocybin-induced changes in suicidality: Oceanic boundlessness and connectedness to nature

OSFNov 26, 2024

Abstract

Psilocybin (4-phosphoryloxyl-N,N-dimethyltryptamine), a natural tryptamine occurring in psilocybe mushrooms, has shown promise in treating a range of mental health conditions, including major depressive disorder (MDD), depression and anxiety in advanced-stage cancer, and alcohol use disorder. There is preliminary evidence from both non-clinical studies of psychedelic use and clinical trials of psilocybin therapy showing reductions in suicidality following psychedelic use. Two closely related concepts – oceanic boundlessness and connectedness to nature – have been proposed as mediators of some of the positive effects of psilocybin. However, their potential role in effects on suicidality has not yet been explored. The Psilocybin in Depression Resistant to standard treatments (PsiDeR) trial was a two arm, double-blind, randomised, placebo-controlled phase 2 trial evaluating feasibility and safety of 25mg psilocybin versus placebo in adults with treatment resistant depression (TRD). This analysis will first assess effects of psilocybin versus placebo on suicidality. The mediating effects of oceanic boundlessness and connectedness to nature will then be explored.
