Psilocybin-Assisted Therapy for Major Depressive Disorder: Perspective from Meta-analysis

OSFNov 1, 2025

Abstract

Six randomized controlled trials (RCTs) have investigated psilocybin in individuals with depression to date, but they revealed inconsistent results. Thus, conclusive evidence remains unavailable to address key clinical questions, including the efficacy and safety of psilocybin for major depressive disorder (MDD), its effect sustainability over time, and optimal dosage constitution. Hence, we will conduct a meta-analysis incorporating the most recent studies, designed to detect even small between-group differences and to assess whether psilocybin produces rapid, sustained antidepressant effects while maintaining safety in individuals with depression.
